Agarose Gel electrophoresis
Gel electrophoresis is a method for separation and analysis of macromolecules (DNA, RNA and proteins) and their fragments, based on their size and charge.
When the charge molecules are placed in an electric field, they migrate towards either the positive or negative pole according to their charge. Nucleic acid molecules are separated by applying an electric field to move the negative charged molecules through an Agarose matrix. Shorter molecules move faster and migrate more easily through the pores of the gel. This phenomenon is called sieving.
2-D Gel Electrophoresis
It is the process for protein separation on the basis of their molecular weight and isoelectric focusing (IEF). This method is more effective than 1-D gel electrophoresis. Separation of protein by isoelectric point is called isoelectric focusing. A protein that is in a pH region below its isoelectric point (pI) will be positively charged and so will migrate towards the cathode (negative). As it migrates through a gradient of increasing pH, however, the protein's overall charge will decrease until the protein reaches the pH region that corresponds to its pI. At this point it has no net charge and so migration ceases (as there is no electrical attraction towards either electrode). As a result, the proteins become focused into sharp stationary bands with each protein positioned at a point in the pH gradient corresponding to its pI. The technique is capable of extremely high resolution with proteins differing by a single charge being fractionated into separate bands.
SDS-PAGE stands for sodium dodecyl sulfate polyacrylamide gel electrophoresis. It is the technique use to separate protein on the basis of their electrophoretic mobility. It means protein is separated on the basis of polypeptide length and charge. Biochemistry, forensics, genetics, molecular biology and biotechnology are some field where this technique is widely used. SDS is an anionic detergent applied to the native protein sample to linearize it and evenly distribute charge per unit mass, thereby resulting in a fractionation by approximate size during electrophoresis.
